The heat released or absorbed at the time of binding is measured directly, and allows for the simultaneous determination of all the thermodynamic parameters of the binding, as well as the affinity and the stoichiometry of the interaction. This measurement is carried out in solution, without prior modification of the partners.<\/p>\n\n<\/div>\n<\/div>\n\n<div class=\"toggler extralight-border\"><span class=\"text-title\"><strong>Description of the equipment<\/strong><\/span><span class=\"toggle_icon extralight-border\"><span class=\"vert_icon extralight-border\"><\/span><span class=\"hor_icon extralight-border\"><\/span><\/span><\/div>\n<div class=\"toggle_wrap\">\n<div class=\"toggle_content\">\n<h3>MicroCal ITC200<\/h3>\n<p>The MicroCal iTC200 provides a detailed overview of the thermodynamic parameters of the binding reaction. The system has a 200 \u03bcl sample cell, and a 50 \u00b5l titration syringe, and allows direct measurement of the heat absorbed or released by mixing precise amounts of reagents during the experiment.<\/p>\n<p>Data analysis is performed using Origin\u2122 software, in which the user obtains the stoichiometry (n), dissociation constant (K<sub>D<\/sub>) and enthalpy (\u0394H) of the interaction.
